Supercontinuum stimulated emission depletion fluorescence lifetime imaging.

چکیده
Supercontinuum (SC) stimulated emission depletion (STED) fluorescence lifetime imaging is demonstrated by using time-correlated single-photon counting (TCSPC) detection. The spatial resolution of the developed STED instrument was measured by imaging monodispersed 40-nm fluorescent beads and then determining their fwhm, and was 36 ± 9 and 40 ± 10 nm in the X and Y coordinates, respectively. The same beads measured by confocal microscopy were 450 ± 50 and 430 ± 30 nm, which is larger than the diffraction limit of light due to underfilling the microscope objective. Underfilling the objective and time gating the signal were necessary to achieve the stated STED spatial resolution. The same fluorescence lifetime (2.0 ± 0.1 ns) was measured for the fluorescent beads by using confocal or STED lifetime imaging. The instrument has been applied to study Alexa Fluor 594-phalloidin labeled F-actin-rich projections with dimensions smaller than the diffraction limit of light in cultured cells. Fluorescence lifetimes of the actin-rich projections range from 2.2 to 2.9 ns as measured by STED lifetime imaging.

Stimulated Emission Depletion (STED) Microscopy: from Theory to Practice
In recent years, various methods have been proposed to increase the resolution of fluorescence microscopy beyond the diffraction limit of light. Amongst them, Stimulated Emission Depletion (STED) microscopy, which relies on a purely physical concept of fluorescence depletion could, in theory, achieve unlimited resolution independent of the wavelength of light. In this chapter, we review the fun...
متن کاملPicosecond polarized supercontinuum generation controlled by intermodal four-wave mixing for fluorescence lifetime imaging microscopy.
We present the generation of a picosecond polarized supercontinuum in highly birefringent multimodal microstructured fiber. The initial steps of the spectral broadening are dominated by intermodal four-wave mixing controlled by the specific fiber design. Using a low repetition rate ultra-stable solid state laser, a pulse train well-suited for versatile time-domain fluorescence lifetime imaging ...
متن کاملphi2FLIM: a technique for alias-free frequency domain fluorescence lifetime imaging.
A new approach to alias-free wide-field fluorescence lifetime imaging in the frequency domain is demonstrated using a supercontinuum source for fluorescence excitation and a phase-modulated image intensifier for detection. This technique is referred to as phi-squared fluorescence lifetime imaging (phi(2)FLIM). The phase modulation and square-wave gating of the image intensifier eliminate aliasi...

Spatial light modulators (SLMs) are a family of optical devices that allow modulation of intensity and/or phase in light microscopy to control excitation and emission. There are broadly two classes of devices employed: digital micromirror arrays (DMDs) and ferroelectric liquid crystal devices. DMDs have found use in controlled light exposure microscopy (CLEM) to reduce photobleaching and photot...
متن کاملAuto-aligning stimulated emission depletion microscope using adaptive optics.
Stimulated emission depletion (STED) microscopy provides diffraction-unlimited resolution in fluorescence microscopy. Imaging at the nanoscale, however, requires precise alignment of the depletion and excitation laser foci of the STED microscope. We demonstrate here that adaptive optics can be implemented to automatically align STED and confocal images with a precision of 4.3 ± 2.3 nm.
